You can do pad assembly or in orbit on any mission but 600-180 ton lift was only one launch

Oops, yeah my bad. I even remember I stacked some fuel tanks and calculated 8×12+4, so I don't even know how I ended with 200 in the end

As long it has a payload door and can land safely it works. STS or X-37B style is your choice.
